Abstract
The present disclosure relates to engineered DNA ligase polypeptides and compositions thereof, as well as polynucleotides encoding the engineered DNA ligase polypeptides. The present disclosure also provides methods of using the engineered DNA ligase polypeptides or compositions thereof in diagnostics and as a molecular biological tool.
Claims13
An engineered DNA ligase, or functional fragment thereof, wherein the DNA ligase or functional fragment thereof has DNA ligase activity and comprises an amino acid sequence sharing at least 85% but less than 100% sequence identity with:
- (a) residues 12 to 437 of SEQ ID NO:2, wherein the amino acid sequence of the engineered DNA ligase comprises one or more substitutions relative to residues 12 to 437 of SEQ ID NO:2, or
- (b) the amino acid sequence of SEQ ID NO:2, wherein the amino acid sequence of the engineered DNA ligase comprises one or more substitutions relative to the amino acid sequence of SEQ ID NO:2,
- wherein the one or more substitutions occur at amino acid position 63, 242, 283, 286, 308, 317, 414, 415, 418, 428, or combinations thereof,
- wherein the amino acid positions are relative to the amino acid sequence of SEQ ID NO:2.
