Abstract
The present disclosure provides methods for treating lung cancer (such as small cell lung cancer, e.g., extensive stage small cell lung cancer) in an individual. The methods comprise administering to the individual a PD-1 axis binding antagonist (such as an anti-PD-L1 antibody, e.g., atezolizumab), a platinum agent (e.g., cisplatin or carboplatin), and a topoisomerase II inhibitor (e.g., etoposide).
Claims35
A method for the treatment of a patient having extensive-stage small cell lung cancer (ES-SCLC), comprising administering to the patient a treatment comprising an effective amount of atezolizumab, an effective amount of carboplatin, and an effective amount of etoposide,
- wherein the atezolizumab, the carboplatin, and the etoposide are each administered in four 21-day dosing cycles (Cycles 1-4), wherein the atezolizumab is administered at a dose of 1200 mg on day 1 of each of Cycles 1-4, the carboplatin is administered at a dose sufficient to achieve AUC=5 mg/ml/min on day 1 of each of Cycles 1-4, and the etoposide is administered at a dose of 100 mg/m2 on each of days 1, 2, and 3 of each of Cycles 1-4, and
- wherein on day 1 of each of Cycles 1-4, the atezolizumab is administered prior to administration of the carboplatin and the etoposide.
A method for the treatment of a patient having ES-SCLC, comprising administering to the patient a treatment comprising an effective amount of atezolizumab, an effective amount of carboplatin, and an effective amount of etoposide,
- wherein the atezolizumab, the carboplatin, and the etoposide are each administered in four 21-day dosing cycles (Cycles 1-4), wherein the atezolizumab is administered at a dose of 1200 mg on day 1 of each of Cycles 1-4, the carboplatin is administered at a dose sufficient to achieve AUC=5 mg/ml/min on day 1 of each of Cycles 1-4, and the etoposide is administered at a dose of 100 mg/m2 on each of days 1, 2, and 3 of each of Cycles 1-4,
- wherein on day 1 of each of Cycles 1-4, the atezolizumab is administered prior to administration of the carboplatin and the etoposide, and
- administering the treatment to patients having ES-SCLC increases the median overall survival (OS) time of the patients that have been administered the treatment as compared to a reference OS time, wherein the reference OS time is the median OS time of patients having ES-SCLC that have been administered a reference treatment comprising carboplatin and etoposide in the absence of atezolizumab, and wherein the OS time of each patient administered the treatment or the reference treatment is measured as the period of time from the start of the treatment or the reference treatment to death of the patient from any cause.
A method of extending OS time of patients having ES-SCLC, the method comprising:
- (a) administering to the patients an effective amount of atezolizumab, an effective amount of carboplatin, and an effective amount of etoposide,
- wherein the atezolizumab, the carboplatin, and the etoposide are each administered in four 21-day dosing cycles (Cycles 1-4), wherein the atezolizumab is administered at a dose of 1200 mg on day 1 of each of Cycles 1-4, the carboplatin is administered at a dose sufficient to achieve AUC=5 mg/ml/min on day 1 of each of Cycles 1-4, and the etoposide is administered at a dose of 100 mg/m2 on each of days 1, 2, and 3 of each of Cycles 1-4,
- wherein on day 1 of each of Cycles 1-4, the atezolizumab is administered prior to administration of the carboplatin and the etoposide, and administering the treatment to patients having ES-SCLC increases the median OS time of the patients that have been administered the treatment as compared to a reference OS time, wherein the reference OS time is the median OS time of patients having ES-SCLC that have been administered a reference treatment comprising carboplatin and etoposide in the absence of atezolizumab, and wherein the OS time of each patient administered the treatment or the reference treatment is measured as the period of time from the start of the treatment or the reference treatment to death of the patient from any cause;
- and
- (b) measuring OS time of the patients to confirm it is extended.
A method for the treatment of a patient having ES-SCLC, comprising administering to the patient a treatment comprising an effective amount of atezolizumab, an effective amount of carboplatin, and an effective amount of etoposide,
- wherein the atezolizumab, the carboplatin, and the etoposide are each administered in four 21-day dosing cycles (Cycles 1-4), wherein the atezolizumab is administered at a dose of 1200 mg on day 1 of each of Cycles 1-4, the carboplatin is administered at a dose sufficient to achieve AUC=5 mg/ml/min on day 1 of each of Cycles 1-4, and the etoposide is administered at a dose of 100 mg/m2 on each of days 1, 2, and 3 of each of Cycles 1-4,
- wherein on day 1 of each of Cycles 1-4, the atezolizumab is administered prior to administration of the carboplatin and the etoposide,
- wherein the atezolizumab is further administered in one or more 21-day dosing cycles following Cycle 4 at a dose of 1200 mg on day 1 of each of the one or more 21-day dosing cycles following Cycle 4, wherein the patients are administered atezolizumab for a duration of at least 12 months in total starting from Cycle 1, and
- administering the treatment to patients having ES-SCLC increases the median OS time of the patients that have been administered the treatment as compared to a reference OS time, wherein the reference OS time is the median OS time of patients having ES-SCLC that have been administered a reference treatment comprising carboplatin and etoposide in the absence of atezolizumab, and wherein the OS time of each patient administered the treatment or the reference treatment is measured as the period of time from the start of the treatment or the reference treatment to death of the patient from any cause.
A method for extending OS time of patients having ES-SCLC, comprising administering to the patients a treatment comprising an effective amount of atezolizumab, an effective amount of carboplatin, and an effective amount of etoposide,
