Abstract
This disclosure relates to compositions and methods of diagnosing neurodegenerative disease by analyzing protein expression profiles in a subject.
Claims20
A method of treating Alzheimer's disease in a subject, said method comprising:
- a) detecting an increased level or a decreased level of at least three signature proteins selected from the group consisting of SUCLG2, NTN1, CIQTNF5, OLFML3, SLIT2, SPON1, GPNMB, SOD2, PRDX3, ALDH6A1, ETFB, HADHA, CYB5R3, CTHRC1, OLFM3, SMOC1, NPTX2, OLFM1, NGEF, DGKG, SLC6A1, GRIN1, APOC2, SDHA, ECI1, HSPA9, and MYL3 in a biological sample from the subject compared to the level of the corresponding signature protein in the control sample, wherein the increased level or the decreased level of the at least three signature proteins in the biological sample relative to the level of the corresponding signature protein from the control sample indicates that the subject has or is at risk of developing Alzheimer's disease, or indicates a progression of Alzheimer's disease in the subject, and
- b) administering to the subject an Alzheimer's disease treatment regimen that is designed configured to modulate increase or decrease the level of at least one of the at least three signature proteins, based upon the level of the at least three signature proteins detected in step a).
A composition comprising one or more reagents for assaying for the level of signature proteins in a sample of a subject, wherein the signature proteins comprise (i) at least six signature proteins selected from the group consisting of ETFB, NPTX2, OLFM1, NGEF, DGKG, SLC6A1, GRIN1, APOC2, SDHA, ECI1, HSPA9, and MYL3, or fragments or variants thereof;
- (ii) at least six signature proteins selected from the group consisting of MAPT, SMOC1, VGF, GPNMB, RPH3A, HSPB1, MDK, CD109, ICAM1, CTHRC1, NTN1, and APP (Aβ) or fragments or variants thereof;
- or
- iii) at least six signature proteins selected from the group consisting of CHI3L1, ENO2, SOD1, MDH1, PGAM1, LDHA, GOT1, GDA, YWHAZ, ALDOA, YWHAG, and PKM or fragments or variants thereof.
A method of treating Alzheimer's disease in a subject, said method comprising:
- a) detecting an increased level or a decreased level of at least one three signature proteins selected from SUCLG2, NTN1, CIQTNF5, OLFML3, SLIT2, SPON1, GPNMB, SOD2, PRDX3, ALDH6A1, ETFB, HADHA, CYB5R3, CTHRC1, OLFM3, SMOC1, NPTX2, OLFM1, NGEF, DGKG, SLC6A1, GRIN1, APOC2, SDHA, ECI1, HSPA9, and MYL3 in a biological sample from the subject, wherein i) the level of two or more of SLIT2, SPON1, GPNMB, CIQTNF5, OLFML3, SMOC1, SUCLG2, PRDX3, NTN1, NPTX2, OLFM1, NGEF, DGKG, SLC6A1, GRIN1, APOC2, SDHA, ECI1, HSPA9, and MYL3 is increased, or ii) the level of two or more of SOD2, PRDX3, ALDH6A1, ETFB, HADHA, CYB5R3, CTHRC1, GFAP, OLFM3, NPTX2, OLFM1, NGEF, DGKG, SLC6A1, GRIN1, APOC2, SDHA, ECI1, HSPA9, and MYL3 is decreased, in the biological sample compared to the level of the corresponding signature protein in a control sample, wherein the increased level or the decreased level of the at least one signature protein in the biological sample relative to the level of the corresponding signature protein from the control sample indicates that the subject has or is at risk of developing Alzheimer's disease, or indicates a progression of Alzheimer's disease in the subject, and
- b) administering to the subject an Alzheimer's disease treatment regimen that reduces the level of at least one of the at least three signature proteins having an increased level or increases the level of at least one of the at least three signature proteins having a decreased level, based upon the level of the at least three signature proteins detected in step a).
