Abstract
Tissue explants of the gastrointestinal tract are provided. Methods of making and using the tissue explants are also provided, along with substrates designed for the tissue explants described.
Claims28
A high-throughput screening composition comprising:
- (i) a first plate comprising a plurality of microwells;
- (ii) a second plate comprising a plurality of microwells, wherein the number of microwells of the second plate matches the number of microwells of the first plate;
- and
- (iii) an isolated tissue explant from a large, non-human, mammalian gastrointestinal tract comprising a luminal surface and a basolateral surface that is placed in between the first plate and the second plate, wherein the luminal surface of the tissue explant is in planar contact with more than one microwell of the first plate and the basolaterial surface of the tissue explant is in planar contact with more than one microwell of the second plate,
- wherein each of the plurality of microwells of the first plate is a through hole that facilitates contact of a test compound with the luminal surface of the tissue explant, and wherein each of the plurality of microwells of the second plate is a receiver chamber capable of containing media that receives the test compound at the basolateral surface of the tissue explant;
- wherein the second plate comprises a solid surface separating each microwell of the plurality of microwells, and wherein the solid surface of the second plate is in planar contact with non-microwell areas of the basolateral surface of the tissue explant;
- and
- wherein the first and second plates apply pressure to non-microwell areas of the tissue explant to minimize well-to-well leakage.
