Abstract
The present invention provides a heparin-insensitive assay for measuring the quantity of Factor XIa in a sample. The present invention provides a method for measuring the concentration of Factor XIa in a plasma sample by using enzymatic heparin degradation as sample pretreatment step.
Claims22
A method of detecting and/or quantifying Factor XIa (FXIa) in a plasma sample containing heparin, comprising:
- a) incubating the plasma sample containing heparin with heparinase, essentially eliminating any heparin-dependent inhibition therein, thereby producing a heparin-depleted sample;
- b) adding a defined amount of Factor IX (FIX) to the heparin-depleted sample and incubating the sample, thereby converting Factor IX to Factor IXa;
- c) adding a defined amount of Factor X (FX) to the sample and incubating the sample, thereby activating Factor Xa in the sample;
- and
- d) detecting and/or quantitating FXIa in the sample by measuring activity of Factor Xa in the heparin-depleted sample.
