A method of modifying a plurality of targeted sites of a double-stranded DNA, comprising a step of introducing a complex comprising a nucleic acid sequence-recognizing module that specifically binds to a target nucleotide sequence in the targeted sites of a double-stranded DNA bonded to Petromyzon marinus cytosine deaminase 1 (PmCDA1) or a nucleic acid encoding the complex, into a mammalian cell containing the double-stranded DNA, and culturing the mammalian cell at a temperature of 20° C. to 25° C. at least temporarily to convert one or more nucleotides in the targeted sites to other one or more nucleotides or delete one or more nucleotides, or insert one or more nucleotides into said targeted sites, without cleaving at least one strand of said double-stranded DNA in the targeted sites,
- wherein the nucleic acid sequence-recognizing module is a clustered regularly interspaced short palindromic repeats (CRISPR)-CRISPR associated (Cas) system comprising a guide RNA complementary to the target nucleotide sequence and a Cas selected from Cas9 and Cpf1, wherein at least one DNA cleavage ability of Cas is inactivated,
- wherein the PmCDA1 has the amino acid sequence set forth in SEQ ID NO:
- 2,
- wherein the PmCDA1 is fused to the nucleic acid sequence-recognizing module by a linker comprising Glycine Serine (GS) linker,
- wherein the complex further comprises a base excision repair inhibitor that is a uracil DNA glycosylase inhibitor (Ugi) linked to PmCDA1, and
- wherein the nucleic acid is an expression vector which is not capable of autonomous replication in the mammalian cell.
