A method of making tirzepatide (SEQ ID NO:1), the method comprising coupling a compound of SEQ ID NO:
- 17 with a compound of SEQ ID NO:20 to form a compound of SEQ ID NO:21.
A compound of SEQ ID NO:
- 21, or a pharmaceutically acceptable salt thereof, wherein one or more selected from the group consisting of Boc, t-Bu, and trt protecting groups may be independently replaced by an alternative protecting group.
