Abstract
The instant disclosure relates to nanoparticle compositions that may be used for the targeting of certain cells or tissues. The nanoparticles may take a variety of different forms, including non-viral, viral, and lipid nanoparticles, and may utilize a TNF receptor superfamily member 12A (“TWEAKR”) binding region of the TWEAK protein to target a nanoparticle to tissues expressing TWEAKR. The compositions may further comprise a suicide gene optionally under the control of a tissue specific promoter. In further aspects, methods of treating an individual using the disclosed nanoparticle compositions are described.
Claims12
A method of treating a glioblastoma, comprising administering to an individual in need thereof a composition comprising a DNA nanoparticle (DNP);
- the DNP comprising
- a 30-mer lysine polymer substituted with polyethylene glycol, wherein the polyethylene glycol has a molecular weight of 10 kDa;
- a protein or peptide ligand having at least 90% sequence identity to a TNF superfamily member 12 (TWEAK) protein and which is specific for TNF receptor superfamily member 12A (“TWEAKR”);
- wherein the protein or peptide ligand is conjugated to the DNP;
- and
- a plasmid comprising a suicide gene under the control of a glioma-cell specific promoter.
