Abstract
Disclosed are methods for treating a cancer selected from breast cancer, colon cancer, ovarian cancer, or pancreatic cancer in a subject in need thereof, comprising administering a therapeutically effective amount of a compound of formula I or a pharmaceutically acceptable salt thereof, in combination with a poly (ADP)-ribose polymerase (PARP) inhibitor. The compound of formula I acts as a checkpoint kinase (CHK) inhibitor. The combination exhibits synergistic anti-tumor efficacy, including enhanced tumor cell growth inhibition at reduced effective concentrations. Also disclosed are pharmaceutically acceptable compositions comprising the compound of formula I and a PARP inhibitor.
Claims16
A method for treating a cancer selected from breast cancer, colon cancer, ovarian cancer, or pancreatic cancer in a subject in need thereof, comprising administering to a subject in need thereof a therapeutically effective amount of a compound of formula I or a pharmaceutically acceptable salt thereof, in combination with a poly(ADP)-ribose polymerase inhibitor,
- wherein Y is NH, O, S, or CH2;
- R1 is selected from the group consisting of:
- wherein X is CH2, NH, S, or O, R8═—H, —NH2, —OH, or —N(R4, R5), wherein R4 and R5, R6are independently H or C1-C6 alkyl,
- R2 is selected from the group consisting of H, OH, NH2, OR14, NR14R15, C1-C6 alkyl, C2-C8 alkenyl, and C2-C8 alkynyl,
- wherein R14 and R15 are independently H or C1-C6 alkyl, and
- R3 is selected from the group consisting of:
- wherein R16, R17, and R18 are independently selected from H, F, Cl, Br, I, and C1-C8 alkyl.
A pharmaceutically acceptable composition, comprising a therapeutically effective amount of a compound of formula I, and a poly (ADP)-ribose polymerase inhibitor,
- wherein Y is NH, O, S, or CH2;
- R1 is selected from the group consisting of:
- wherein X is CH2, NH, S, or O,
- R8═—H, —NH2, —OH, or —N(R4, R5), wherein R4 and R5 are independently H or C1-C6 alkyl,
- R2 is selected from the group consisting of H, OH, NH2, OR14, NR14R15, C1-C6 alkyl, C2-C8 alkenyl, and C2-C8 alkynyl, wherein R14 and R15 are independently H or C1-C6 alkyl, and
- R3 is selected from the group consisting of:
- wherein R16, R17, and R18 are independently selected from H, F, Cl, Br, I, and C1-C8 alkyl.
