Abstract
The present invention relates to novel tetrazine compounds of formula I, wherein one of R1-R5 is 18F, for use in pretargeted in vivo imaging. The compounds are suitable for use in click chemistry, i.e. reactions that join a targeting molecule and a reporter molecule. The invention further relates to precursors to formula I, wherein one of R1-R5 is SnR3, B(OR)2, B(0H)2. Formula (I).

A tetrazine compound comprising a tetrazine moiety and having the following Formula I:
- wherein:
- one of R1-R5 is 18F,
- at least two of the remaining R1-R5 are H, and the other remaining R1-R5 are the same or different and are selected from H, alkyl, halogen, —CF3, —CN, —O-alkyl, —S-alkyl, —NH-alkyl, —N(alkyl)2, —NH(C═O)-alkyl, —N-alkyl-(C═O)-alkyl, —SO2-alkyl, —SO2—NH2, —SO2—NHalkyl, —SO2—N(alkyl)2-C(═O)—NH2, —C(═O)—NH-alkyl, —C(═O)—N(alkyl)2, —C(═O)—OH, —C(═O)—O-alkyl, —CH2—NH2, —CH2—NH-alkyl, —OH, CH2—O-alkyl, CH2—O-aryl, CH2—O-phenyl, CH2—O-naphthyl, and
- wherein n is an integer from 1 to 4, and
- R6 is selected from H, CH3, unsubstituted phenyl,
- wherein the curly bond indicates a link to the tetrazine moiety.
A tetrazine compound comprising a tetrazine moiety and having the following Formula I:
- wherein:
- R2 or R4 is 18F,
- at least two of R1-R5 are H, and the other remaining R1-R5 are the same or different and are selected from H, alkyl, halogen, —CF3, —CN, —O—alkyl, —S—alkyl, —NH—alkyl, N(alkyl)2, —NH(C═O)—alkyl, —N—alkyl—(C═O)—alkyl, —SO2—alkyl, —SO2—NH2, —SO2—NHalkyl, —SO2—N(alkyl)2—C(═O)—NH2, —C(═O)—NH—alkyl, —C(═O)—N(alkyl)2, —C(═O)—OH, —C(═O)—O—alkyl, —CH2—NH2, —CH2—NH—alkyl, —OH, CH2—O—alkyl, CH2—O—aryl, CH2—O—phenyl, CH2—O—naphthyl, and
- wherein n is an integer from 1 to 4, and
- R6 is selected from H, CH3, phenyl,
- wherein the curly bond indicates a link to the tetrazine moiety.
A tetrazine compound comprising a tetrazine moiety and having the following Formula II:
- wherein:
- R7 is selected from —CH2—NH2, —CH2—NH—alkyl, —C(═O)—NH2, —C(═O)NH—alkyl, —NH—CH(═O), —NH—C(═O)—alkyl, —OH, —O—alkyl, —CH2—O—alkyl, —CH2—O—phenyl, —CH2—O—naphthalene, —CH2—C(═O)NH2, and —CH2—C(═O)—NH—alkyl.
