Abstract
This invention relates to compositions, methods, strategies, and treatment modalities related to the epigenetic modification of hepatitis B virus (HBV) genes.
Claims19
A method, comprising administering an epigenetic editing system to a subject, wherein the subject comprises detectable levels of HBV DNA, HBsAg, and/or HBeAg in plasma of the subject, wherein the epigenetic editing system comprises:
- a) a fusion protein comprising
- i) a dCas9 protein domain,
- ii) a DNMT3A domain, and
- iii) a human KRAB domain,
- or one or more nucleic acid molecules encoding the fusion protein, and
- b) a guide RNA (gRNA) consisting of
- i) a targeting domain, consisting of a targeting sequence that is 100% complementary to a target sequence, wherein the target sequence consists of the sequence of SEQ ID NO:
- 391, and
- ii) a Cas9 tracrRNA.
A method, comprising administering an epigenetic editing system to a subject, wherein the subject comprises detectable levels of HBV DNA, HBsAg, and/or HBeAg in plasma of the subject, wherein the epigenetic editing system comprises:
- a) a fusion protein comprising
- i) a dCas9 protein domain,
- ii) a DNMT3A domain, and
- iii) a human KRAB domain, or one or more nucleic acid molecules encoding the fusion protein, and
- b) a guide RNA (gRNA) consisting of
- i) a targeting domain, consisting of a targeting sequence that is 100% complementary to a target sequence, wherein the target sequence consists of the sequence of SEQ ID NO:
- 391, and
- ii) a tracrRNA consisting of a sequence comprising the sequence of SEQ ID NO:
- 1089.
- 391, and
- ii) a tracrRNA consisting of a sequence comprising the sequence of SEQ ID NO:
- 1087.
