Claims19
A sequencing method, comprising:
- providing, in a nucleic acid sequencing reaction, an oligonucleotide and a combination of reversibly terminated nucleotides and non-labeled nucleotides that are not reversibly terminated;
- and
- performing the nucleic acid sequencing reaction, wherein nucleotides of the oligonucleotide that correspond with the reversibly terminated nucleotides are sequenced, wherein nucleotides of the oligonucleotide that correspond with the non-labeled nucleotides that are not reversibly terminated are not sequenced, and wherein (a) or (b) is true:
- (a) the oligonucleotide comprises a non-sequenced region that includes only a subset of nucleotides selected from A, C, G, and T, wherein the subset is not sequenced, and wherein the non-sequenced region is hybridizable to a recode nucleic acid sequence indicative of an amino acid identity, or is hybridizable to a cycle nucleic acid sequence indicative of a workflow cycle;
- or
- (b) the oligonucleotide comprises a sequenced region comprising a recode nucleic acid sequence indicative of an amino acid identity and a cycle nucleic acid sequence indicative of a workflow cycle, and wherein the oligonucleotide further comprises a memory oligonucleotide comprising the recode nucleic acid sequence and the cycle nucleic acid sequence.
