Provided is a humanized anti-IL-4Rα single domain antibody. The antibody has complementarity determining regions and humanized and modified framework regions.

A humanized anti-IL-4Rα single domain antibody, comprising complementarity determining regions comprising CDR1, CDR2 and CDR3 and framework regions comprising FR1, FR2, FR3 and FR4;
- wherein CDR1 has an amino acid sequence as set forth in SEQ ID NO. 3, CDR2 has an amino acid sequence as set forth in SEQ ID NO. 12, and CDR3 has an amino acid sequence as set forth in SEQ ID NO. 18;
- and
- FR1 has an amino acid sequence as set forth in SEQ ID NO. 2, FR2 has an amino acid sequence selected from any one of SEQ ID NO. 4-11, FR3 has an amino acid sequence selected from any one of SEQ ID NO. 13-17, and FR4 has an amino acid sequence as set forth in SEQ ID NO. 20 or 21.
