Abstract
The present invention provides humanized monoclonal antibodies that recognize human Nectin-2 with high affinity and specificity and inhibit its binding to CD112R. These antibodies carry cytotoxic payload and do not interact with Fc gamma receptors. The present invention further provides pharmaceutical compositions comprising the antibodies and methods for their use in cancer immunotherapy.
Claims6
A conjugate, comprising:
- (a) an antibody that binds Nectin-2 or an antigen-binding fragment thereof that comprises:
- A) a heavy chain variable region (VH) comprising an amino acid sequence at least 90% identical to the amino acid sequence set forth in SEQ ID NO:
- 11 and (i) a complementarity determining region (CDR) 1 having an amino acid sequence according to SEQ ID NO:
- 1;
- (ii) a CDR2 having an amino acid sequence according to SEQ ID NO:
- 2;
- and (iii) a CDR3 having an amino acid sequence according to SEQ ID NO:
- 3;
- and
- B) a light chain variable region (VL) comprising an amino acid sequence at least 90% identical to the amino acid sequence set forth in SEQ ID NO:
- 12 and (i) a CDR1 having an amino acid sequence according to SEQ ID NO:
- 4;
- (ii) a CDR2 having an amino acid sequence according to SEQ ID NO:
- 5;
- and (iii) a CDR3 having an amino acid sequence according to SEQ ID NO:
- 6;
- and
- (b) a topoisomerase I inhibitor conjugated to the Nectin-2 antibody or antigen-binding fragment thereof using a linker.
