Abstract
The present disclosure discloses a class of nitrogen-containing fused heterocyclic compounds of N-sulfonamide and the use thereof. The nitrogen-containing fused heterocyclic compounds of N-sulfonamide of the present disclosure are compounds of formula I, isotope-labeled compounds thereof, solvates thereof, pharmaceutically acceptable salts thereof, solvates of the pharmaceutically acceptable salts thereof. Such compounds can promote the binding of p53 Y220C mutant with DNA and possess better inhibitory activity against the proliferation of cancer cells.

A nitrogen-containing compound of formula I, an isotope-labeled compound thereof, a solvate thereof, a pharmaceutically acceptable salt thereof, or a solvate of the pharmaceutically acceptable salt thereof;
- wherein
- X1, X2, and X3 are each independently selected from CH and N;
- R1, R2, R3, R4, and R5 are each independently selected from hydrogen and deuterium;
- R6 is —(CH2)nR7;
- n is 0, 1, 2, or 3;
- R7 is selected from C1-C6 alkyl, deuterated C1-C6 alkyl, C1-C6 haloalkyl, and C3-C6 cycloalkyl.
A compound of formula a-11 or a-32;
- wherein X is halogen;
- R8 is an amino protecting group;
- X1, X2, and X3 are each independently selected from CH and N;
- R1, R2, R3, R4, and R5 are each independently selected from hydrogen and deuterium;
- R6 is —(CH2)nR7,
- n is 0, 1, 2, or 3;
- R7 is selected from C1-C6 alkyl, deuterated C1-C6 alkyl, C1-C6 haloalkyl, and C3-C6 cycloalkyl.
