Abstract
The invention relates to PGT121-germline-targeting designs, trimer stabilization designs, combinations of those two, trimers designed with modified surfaces helpful for immunization regimens, other trimer modifications and on development of trimer nanoparticles and methods of making and using the same.
Claims12
A non-naturally occurring protein comprising an immunogen, wherein the immunogen is a human immunodeficiency virus type 1 (HIV-1) gp140 trimer derived from BG505 isolate sequence, and wherein each protomer of said gp140 trimer comprises MD39 mutations, wherein the MD39 mutations comprise N130A, L165C, A73C, I290C, M162C, A316V, N279K, A501C, T567C, I544V, and T605V, and wherein said MD39 mutations confer improved thermostability and a native-like antigenic profile as compared to a corresponding gp140 trimer lacking said mutations.
