Abstract
The present disclosure provides compositions and methods for polynucleotide synthesis in vitro, specifically the use of single-subunit RNA polymerases for synthesis and manufacturing of RNA.
Claims9
A method of performing in vitro transcription comprising the steps of
- (a) combining
- (i) a polypeptide having at least 95% amino acid sequence identity to a single-subunit RNA polymerase (RNApol) molecule selected from the group consisting of SEQ ID NOs:
- 21-25, and 62-66,
- (ii) a double-stranded DNA template comprising a promoter selected from the group consisting of SEQ ID NOs:
- 103-107 operatively linked to a polynucleotide encoding a protein or polypeptide;
- and
- (iii) one or more nucleotide triphosphates;
- (b) incubating the combination of (a) at a temperature from 10° C. to 80° C.;
- and
- (c) producing a transcript or a population of transcripts.
