Dry powder formulations for messenger RNA

A method for manufacturing a dry powder formulation comprising a plurality of spray-dried particles, the method comprising:

• spray-drying a mixture comprising one or more messenger RNAs (mRNAs), one or more lipids, and one or more polymers to form the plurality of spray-dried particles, wherein:
• the one or more polymers comprise a polymethacrylate based polymer;
• a moisture content of the plurality of spray-dried particles is less than 10%;
• the plurality of spray-dried particles comprises mRNA-loaded nanoparticles;
• and
• the mRNA-loaded nanoparticles have an mRNA encapsulation efficiency of 75% or greater after spray-drying the mixture.
