Abstract
Use of a TGF-β small molecule inhibitor in the field of nerve regeneration, for in vitro regeneration and directed differentiation of a variety of nerve cells and brain-like organs. By adding the TGF-β small molecule inhibitor to a set of basal media having clear chemical compositions, pluripotent stem cells can be induced into adult cells derived from a variety of neural stem cells, and the number of induced nerve cells can be greatly increased.
Claims13
An inducing medium for neural stem cells and organoids, wherein the inducing medium consists of basal medium and 10 nM-100 μM TGF-β inhibitor;
- the basal medium consists of Dulbecco's Modified Eagle/F12 Medium, 1% minimum essential medium non-essential amino acids, 0.5 g/L sodium chloride, 13.6 μg/L sodium selenite, 22 μg/mL insulin and 100 ng/mL recombinant human transferrin;
- wherein the TGF-β inhibitor is 4-[2-(6-methylpyridin-2-yl)-5,6-dihydro-4H-pyrrolo[1,2-b]pyrazol-3-yl]quinoline-6-carboxamide.
