Abstract
Disclosed herein are methods of diagnosing and treating MYC-driven cancers by detecting a PVT1 splice variant in a biological sample from a subject.

A method of treating a cancer in a subject, the method comprising administering to the subject a therapy targeting a PVT1 splice variant CircPVT1, wherein the therapy comprises an siRNA directed to the PVT1 splice variant CircPVT1, wherein the siRNA comprises a nucleotide sequence that is complementary to SEQ ID NO. 1 or a portion thereof, wherein the siRNA comprises the nucleotide sequence set forth in SEQ ID NO. 3 or SEQ ID NO. 4, and wherein the subject has previously been identified as having a cancer characterized by a co-gain of a PVT1 and a c-Myc.
