Abstract
The present disclosure is directed to compounds of formulas (I)-(VI), which are useful as modulators of USP36. The compounds are further useful in the inhibition of USP36 and the treatment of diseases or disorders associated with the inhibition of USP36. For instance, the disclosure is concerned with compounds and compositions for inhibition of USP36, methods of treating diseases associated with the inhibition of USP36 (e.g., certain forms of cancer), and methods of synthesis of these compounds.

A compound of formula (II):
- or a pharmaceutically acceptable salt thereof, wherein:
- Y is N or CH;
- R1 is selected from:
- (C1-C4) alkyl optionally substituted with 1-3 R2,
- (C3-C6) cycloalkyl optionally substituted with one R3,
- aryl optionally substituted with 1-2 R4,
- heteroaryl substituted with one R5,
- bicyclic heteroaryl optionally substituted with one (C1-C4) alkyl,
- partially saturated bicyclyl optionally substituted with one halogen, and
- each R2 is independently selected from:
- (C1-C4) alkyl,
- (C3-C6) cycloalkyl,
- (C6-C12) spirocycloalkyl,
- aryl optionally substituted with one halogen or —OR6, and
- 3-6 membered heterocyclyl;
- R3 is (C1-C4) alkyl;
- each R4 is independently selected from (C1-C4) alkyl optionally substituted with halogen, —OR6, halogen, and 3-6 membered heterocyclyl;
- R5 is aryl substituted with one halogen;
- R6 is selected from aryl, and (C1-C4) alkyl optionally substituted with halogen;
- R10 is hydrogen or halogen;
- and
- R11 is halogen or (C1-C4) alkyl substituted with halogen.
