Abstract
The present disclosure is directed to compounds of formulas (I)-(VI), which are useful as modulators of USP19. The compounds are further useful in the inhibition of USP19 and the treatment of diseases or disorders associated with the inhibition of USP19. For instance, the disclosure is concerned with compounds and compositions for inhibition of USP19 and methods of treating diseases associated with the inhibition of USP19 (e.g., Parkinson's disease, Ewing sarcoma, and other metabolic diseases including muscle wasting and diabetes).
Claims12
A compound of formula (I):
- and enantiomers, diastereomers, and pharmaceutically acceptable salts thereof, wherein:
- X is N or C;
- wherein if X is N, one of R1 and R1′ is absent and one of R2 and R2′ is absent;
- Y is N or CR7;
- Z is CH or CH2;
- R1 and R1′ are each independently selected from H, C1-4 alkyl, and aryl optionally substituted with one R8;
- R2 and R2′ are each independently selected from H and C1-4 alkyl;
- wherein one of R1 or R1′ may be joined to one of R2 or R2′ to form an aryl or heteroaryl ring that includes the atoms to which they are attached and is substituted with one R9;
- one of R1 or R1′ and one of R2 or R2′ may be absent, such that ring A contains an unsaturated bond between X and the carbon to which R2 and R2′ are connected;
- R3 is selected from:
- H, —OR10, and halogen;
- R4 is H or C1-4 alkyl;
- R5 is H or C1-4 alkyl;
- wherein R4 and R5 may be joined to one another to form a cycloalkyl that includes the carbon to which they are attached;
- R6 is selected from:
- alkylcycloalkyl, alkylaryl, and alkylheteroaryl, each of which may optionally be substituted with one C1-4 alkyl;
- R7 is —C(O)R11;
- R8 is selected from:
- halogen, C1-4 alkyl, and C1-4 alkoxy;
- R9 is selected from:
- H, halogen, C1-4 alkyl, C1-4 alkoxy, —C(O)N(R12)2, and heteroaryl;
- R10 is H or C1-4 alkyl;
- R11 is —N(R12)2 or a 5 to 6-membered heterocycle containing two heteroatoms selected from N and O;
- R12 is a C1-4 alkyl group;
- and
- m is 0.
