Abstract
A method of sequencing a target nucleic acid polymer by (a) modifying a target nucleic acid polymer to produce a modified nucleic acid polymer; (b) producing fragments of the modified nucleic acid polymer, wherein the fragments are attached to locations on a solid support surface (c) determining nucleotide sequences from the fragments at the locations; and (d) producing a representation of the nucleotide sequence for the target nucleic acid polymer based on the nucleotide sequences from the fragments and the relative distances between the locations on the solid support surface.
Claims21
A method for analyzing target nucleic acids, comprising:
- fragmenting target nucleic acids to form polynucleotide fragments;
- capturing the polynucleotide fragments on a solid support surface, wherein polynucleotide fragments produced from a particular target nucleic acid molecule are localized to a respective region on the solid support surface;
- determining proximity of the polynucleotide fragments from each other on the solid support surface by evaluating relative locations of the polynucleotide fragments on the solid support surface;
- and
- determining which polynucleotide fragments are derived from a common target nucleic acid molecule based on the proximity of the polynucleotide fragments on the solid support surface.
