Abstract
Methods of identifying a sequence of a polypeptide within a heterogenous mixture of polypeptides, the polypeptide being immobilized to a support and having at least one labeled amino acid residue. Methods involve detecting at least one signal or signal change from the immobilized polypeptide and subjecting the polypeptide to conditions sufficient to remove at least one amino acid residue from the polypeptide.
Claims21
A method for analyzing a polypeptide, comprising:
- (a) providing said polypeptide immobilized to a support, wherein said polypeptide comprises at least one labeled internal amino acid residue;
- (b) identifying at least a portion of a sequence of said;
- and
- (c) subjecting said polypeptide to conditions sufficient to remove at least one amino acid residue from said polypeptide;
- wherein said identifying at least a portion of a sequence of said polypeptide comprises detecting at least one signal or signal change from said polypeptide immobilized to said support and comparing the detected at least one signal or signal change to a simulated signal or signal change corresponding to a known polypeptide sequence, wherein the simulated signal or signal change is generated by a method comprising a step of simulating at least one modeled experimental error of a single-molecule sequencing method.
