Abstract
Methods of modulating expression of a target nucleic acid in a cell are provided including use of multiple orthogonal Cas9 proteins to simultaneously and independently regulate corresponding genes or simultaneously and independently edit corresponding genes.
Claims14
A method of altering two or more target nucleic acids in a cell comprising
- forming within the cell two or more co-localization complexes, with each including a guide RNA, an orthogonal RNA guided DNA binding protein of the Type II CRISPR system and a target nucleic acid and wherein each of the orthogonal RNA guided DNA binding protein of the Type II CRISPR system of the two or more co-localization complexes is orthogonal to each other, and cuts or nicks the target nucleic acid.
