Abstract
Compositions and methods for the simulataneous capture and release using micropattern surfaces for tissue and cell microdissection. In one example, a patterned thermoplastic film has a first surface and a plurality of projections attached to and extending outwardly from the first surface. The projections form a pattern on the thermoplastic film.
Claims17
A method of isolating one or more portions of a tissue of interest from a tissue sample, the method comprising:
- (a) positioning the tissue sample on its first surface on a first side of a glass slide that is coated with non-adhesive microparticles or nanoparticles;
- (b) contacting an opposing second surface of the tissue sample with a face of a patterned thermoplastic film, wherein the face of the patterned thermoplastic film comprises one or more projections attached to and extending outwardly from said face of the patterned thermoplastic film;
- (c) identifying the one or more portions of the tissue of interest in the tissue sample;
- (d) irradiating the patterned thermoplastic film in contact with the tissue sample with a radiation comprising an electromagnetic energy, a first wavelengths in an UV spectrum, or a second wavelength in an IR spectrum,
- wherein irradiation of the patterned thermoplastic film causes the patterned thermoplastic film to melt onto a contacting surface of the tissue sample above the one or more portions of the tissue of interest and wherein melting the patterned thermoplastic film onto the contacting surface of the tissue sample causes one or more portions of the tissue of interest to adhere to the patterned thermoplastic film;
- and
- (e) removing the patterned thermoplastic film and the one or more portions of the tissue of interest adhering to the patterned thermoplastic film from remainder of the tissue sample;
- and
- wherein the one or more projections are impregnated with a gradient of an UV absorbing dye or an IR absorbing dye, and wherein the one or more projections impregnated with the gradient of the UV absorbing dye or the IR absorbing dye swell to conform to surface irregularities of the tissue sample upon irradiation with the radiation.
A method of isolating one or more portions of a tissue of interest from a tissue sample, the method comprising:
- (a) positioning the tissue sample on its first surface on a first side of a glass slide that is coated with non-adhesive microparticles or nanoparticles;
- (b) contacting an opposing second surface of the tissue sample with a face of a patterned thermoplastic film, wherein the face of the patterned thermoplastic film comprises one or more projections attached to and extending outwardly from said face of the patterned thermoplastic film;
- (c) identifying the one or more portions of the tissue of interest in the tissue sample;
- (d) irradiating the patterned thermoplastic film in contact with the tissue sample with a radiation comprising an electromagnetic energy, a first wavelength in an UV spectrum, or a second wavelength in an IR spectrum,
- wherein irradiation of the patterned thermoplastic film causes the patterned thermoplastic film to melt onto a contacting surface of the tissue sample above the one or more portions of the tissue of interest and wherein melting the patterned thermoplastic film onto the contacting surface of the tissue sample causes one or more portions of the tissue of interest to adhere to the patterned thermoplastic film;
- and
- (e) removing the patterned thermoplastic film and the one or more portions of the tissue of interest adhering to the patterned thermoplastic film from remainder of the tissue sample;
- and
- wherein each of the one or more projections defines at least one tip, and wherein at least one tip of at least one of the one or more projections is coated with one or more particles, or wherein each of the one or more projections comprises a microneedle, a microsphere, or a micropillar.
A system for isolating one or more portions of a tissue of interest from a tissue sample via a laser capture microdissection, the system comprising:
- (a) a glass slide with a non-adhesive coating on a first side of the glass slide;
- (b) a tissue sample positioned on the first side of the glass slide on a first surface of the tissue sample;
- (c) a micropatterned thermoplastic film, wherein a face of the micropatterned thermoplastic film comprises one or more projections attached to and extending outwardly from said face of the micropatterned thermoplastic film and wherein the face of the micropatterned thermoplastic film is in contact with an opposing second surface of the tissue sample, wherein the one or more projections are impregnated with a gradient of an UV absorbing dye or an IR absorbing dye, and wherein the one or more projections impregnated with the gradient of the UV absorbing dye or the IR absorbing dye swell to conform to surface irregularities of the tissue sample upon irradiation with a radiation comprising an electromagnetic energy, a first wavelength in an UV spectrum, or a second wavelength in an IR spectrum;
- and
- (d) a laser capture microdissection instrument configured to irradiate the tissue through an opposing second side of the micropatterned thermoplastic film with the radiation.
A system for isolating one or more portions of a tissue of interest from a tissue sample via a laser capture microdissection, the system comprising:
- (a) a glass slide with a non-adhesive coating on a first side of the glass slide;
- (b) a tissue sample positioned on the first side of the glass slide on a first surface of the tissue sample;
- (c) a micropatterned thermoplastic film, wherein a face of the micropatterned thermoplastic film comprises one or more projections attached to and extending outwardly from said face of the micropatterned thermoplastic film and wherein the face of the micropatterned thermoplastic film is in contact with an opposing second surface of the tissue sample;
- and
- (d) a laser capture microdissection instrument configured to irradiate the tissue through an opposing second side of the micropatterned thermoplastic film with a radiation comprising an electromagnetic energy, a first wavelength in an UV spectrum, or a second wavelength in an IR spectrum;
- and wherein the one or more projections defines at least one tip, and wherein at least one tip of at least one of the one or more projections is coated with one or more particles.
