Abstract
A promoter operably linked to a gene encoding a protein is disclosed. The promoter drives expression of the protein in a yeast cell in the absence of methanol. Also disclosed are vectors, host cells and expression systems that include the promoter, as well as methods of using the promoter to express proteins in yeast.
Claims29
An isolated nucleic acid sequence, having at least 95% sequence identity to at least 1150 residues of any one of SEQ ID NOs:
- 1-6 operably linked to a gene encoding a protein heterologous to Komagataella phaffii, and the nucleic acid sequence drives expression of the protein in a yeast cell grown in the absence of methanol, wherein the yeast cell is of the genus Komagataella.
A method of expressing a protein in a yeast cell comprising:
- providing a yeast cell;
- introducing an isolated nucleic acid sequence into the yeast cell,
- the isolated nucleic acid sequence being operably linked to a gene encoding the protein,
- the isolated nucleic acid sequence having at least 95% sequence identity to at least 1150 residues of any one of SEQ ID NOs:
- 1-6;
- fermenting the yeast cell under at least one fermentation condition in the absence of methanol in a nutrient broth;
- harvesting the yeast cell;
- and
- recovering the protein from the yeast cell,
- wherein the protein is heterologous to Komagataella phaffii, and
- wherein the yeast cell is of the genus Komagataella.
