Abstract
The disclosure provides a chimeric antigen receptor (CAR) comprising a modified hinge, transmembrane and/or intracellular domain disclosed herein. Some aspects of the disclosure relate to a polynucleotide encoding a chimeric antigen receptor (CAR) comprising the costimulatory domain disclosed herein. Other aspects of the disclosure relate to cells comprising the CAR and use in a T cell therapy.

A vector encoding a polypeptide comprising a chimeric antigen receptor (CAR), wherein the CAR comprises:
- (i) an antibody or antigen-binding domain selected from the group consisting of a single chain fragment (scFv), an affibody, a Fab fragment, a F(ab′)2 fragment, and a disulfide-linked Fv (sdFv);
- (ii) a transmembrane domain comprising the amino acid sequence of EITLIIFGVMAIVIGTILLISYGI (SEQ ID NO:69) or LTSIISAVVVILLVVVLGVVFGILIK (SEQ ID NO:
- 70);
- and
- (iii) an intracellular domain.
