Abstract
The present invention relates to the field of biochemistry, more particularly to proteomics, more particularly to protein sequencing, even more particularly to single molecule peptide sequencing. The invention discloses methods for single molecule protein sequencing and/or amino acid identification using cleavage inducing agents which are not specific for one particular amino acid, cleave polypeptides step by step from the N-terminus onwards and provide information on the identity of the cleaved amino acids based on the reaction kinetics.
Claims19
A method of detecting terminal amino acid cleavage, the method comprising:
- a) contacting an immobilized peptide with one or more peptide-binding agents;
- b) measuring residence times of the one or more peptide-binding agents on the immobilized peptide, wherein each residence time corresponds to a single molecule binding event between a peptide-binding agent and a terminal amino acid of the immobilized peptide;
- and
- c) detecting one or more changes in the measured residence times, wherein a change in the measured residence times is indicative of terminal amino acid cleavage from the immobilized peptide.
