Abstract
Disclosed are compositions and methods for decreasing the viscosity and/or cohesiveness of and/or increasing the liquefaction of excessively or abnormally viscous or cohesive mucus or sputum. The composition contains a protein or peptide containing a thioredoxin monocysteinic active site in a reduced state and optionally further contains a reducing system.
Claims14
A method to treat a respiratory condition in a patient,
- comprising administering a composition comprising a protein or peptide containing a thioredoxin monocysteinic active site in a reduced state to the respiratory tract of the patient, wherein the thioredoxin monocysteinic active site comprises an amino acid A sequence selected from the group consisting of C-X-X-S(SEQ ID NO:
- 24), X-C-G-P-X-X (SEQ ID NO:21), W-C-G-P-X-K (SEQ ID NO:23), X-C-X-X-S-X (SEQ ID NO:25), X-C-G-P-S-X (SEQ ID NO:26), and W-C-G-P-S-K (SEQ ID NO:27), wherein the C residue is in a reduced state, and wherein the X residues are any amino acid residue other than 2 cysteine.
A method to treat a disease selected from the group consisting of cystic fibrosis, chronic obstructive pulmonary disease, bronchiectasis, and asthma in a patient, comprising administering to the patient a pharmaceutical composition comprising a protein or peptide containing a thioredoxin monocysteinic active site in a reduced state, wherein the composition is formulated for oral administration, topical administration, or aerosol administration to the lung, wherein the thioredoxin monocysteinic active site comprises an amino acid sequence selected from the group consisting of C-X-X-S(SEQ ID NO:
- 24), X-C-G-P-X-X (SEQ ID NO:21), W-C-G-P-X-K (SEQ ID NO:23), X-C-X-X-S-X (SEQ ID NO:25), X-C-G-P-S-X (SEQ ID NO:26), and W-C-G-P-S-K (SEQ ID NO:27), wherein the C residue is in a reduced state, and wherein the X residues are any amino acid residue other than cysteine.
