Abstract
Provided herein are methods for capturing an analyte from a first region of interest of a biological sample on a substrate, where the biological sample comprises the first region of interest and a second region, and where the method includes contacting the second region with a sealant in order to create a hydrophobic seal thereby preventing an interaction between an analyte from the second region with a capture domain of a capture probe.
Claims18
A method for capturing an analyte from a region of interest of a tissue sample, the method comprising:
- (a) contacting the tissue sample with an array such that the tissue sample is aligned with the array, wherein the array comprises a plurality of capture probes, wherein each capture probe of the plurality of capture probes comprises a spatial barcode and a capture domain,
- wherein the tissue sample comprises:
- (i) the region of interest, wherein the region of interest is aligned with a first capture probe of the plurality of capture probes;
- and
- (ii) a region of non-interest, wherein the region of non-interest is aligned with a second capture probe of the plurality of capture probes;
- (b) selectively contacting the region of non-interest of the tissue sample with a liquid coverslip sealant, wherein the liquid coverslip sealant prevents permeabilization of the region of non-interest and hybridization of an analyte from the region of non-interest to the second capture probe of the plurality of capture probes;
- and
- (c) hybridizing an analyte from the region of interest to the capture domain of the first capture probe, thereby capturing the analyte from the region of interest of the tissue sample.
