Abstract
Disclosed herein are novel compounds, pharmaceutical compositions comprising such compounds and related methods of their use. The compounds described herein are useful, e.g., as liposomal delivery vehicles to facilitate the delivery of encapsulated polynucleotides to target cells and subsequent transfection of said target cells, and in certain embodiments are characterized as having one or more properties that afford such compounds advantages relative to other similarly classified lipids.

A lipid nanoparticle comprising:
- mRNA;
- one or more helper lipids, one or more non-cationic lipids, and/or one or more PEG-modified lipids;
- and
- a cationic lipid having the structure:
- wherein
- R1 and R2 are each independently an optionally substituted saturated C1-C20 alkyl or an optionally substituted variably unsaturated C2-C20 alkenyl
- L1 is selected from the group consisting of hydrogen, an optionally substituted C3-C30 alkyl, and an optionally substituted variably unsaturated C3-C30 alkenyl;
- L2 is selected from the group consisting of an optionally substituted C3-C30 alkyl and an optionally substituted variably unsaturated C3-C30 alkenyl;
- m is three, four, five, six, seven, eight, nine, ten, eleven, twelve, thirteen, fourteen, fifteen, sixteen, seventeen, eighteen, nineteen, or twenty;
- o is zero, one, two, three, four, five, six, seven, eight, nine, ten, eleven, twelve, thirteen, fourteen, fifteen, sixteen, seventeen, eighteen, nineteen, or twenty;
- and
- n is zero, one, two, three, four, five, six, seven, eight, nine, ten, eleven, twelve, thirteen, fourteen, fifteen, sixteen, seventeen, eighteen, nineteen, or twenty.
