Replicating and non-replicating vectors for recombinant protein production in plants and method of use thereof

A plant expression vector comprising an expression cassette, wherein the expression cassette comprises a 5′ UTR and a 3′ UTR operably linked to a nucleotide sequence encoding a protein of interest, short intergenic region of bean yellow dwarf virus (SIR), and p19 suppresser of RNA silencing from tomato bushy stunt virus (P19);

• wherein the 3′ UTR comprises:
• a first terminator, wherein the first terminator is intronless tobacco extension terminator (EU);
• and
• a second terminator, wherein the second terminator is from a 3′ UTR selected from the group consisting of:
• Nicotiana benthamiana actin 3′ UTR (NbACT3), [p19 suppressor of RNA silencing from tomato bushy stunt virus (P19),] N. benthamiana 18.8 kDa class II heat shock protein 3′ UTR (NbHSP), agrobacterium nopaline synthase 3′ UTR (NOS), cauliflower mosaic virus 35S 3′ UTR (35S), tobacco mosaic virus 3′ UTR (TMV), BDBS501 (bean dwarf mosaic virus DNA B nuclear shuttle protein 3′ UTR, the intergenic region, the 3′ end of the movement protein, and additional 200 nt downstream of the movement protein sequence), tobacco necrosis virus-D 3′ UTR (TNVD), pea enation mosaic virus 3′ UTR (PEMV), and barley yellow dwarf virus 3′ UTR (BYDV);
• wherein the first terminator and second terminator are operably linked.
