Controlled-release and stratified cyclodextrin inclusion complex vehicles

A method of formulating a composition for oral delivery to a human subject in need thereof, the method comprising:

• providing a cyclodextrin having a cavity, wherein the cyclodextrin is an alpha-cyclodextrin, a beta-cyclodextrin, or a gamma-cyclodextrin;
• admixing the cyclodextrin with N-acetylcysteine and acetaminophen so that N-acetylcysteine and acetaminophen are retained as stacked guest molecules within the cavity of the cyclodextrin, forming a cyclodextrin inclusion complex;
• admixing the cyclodextrin inclusion complex with a pharmaceutically acceptable carrier for oral delivery, wherein the N-acetylcysteine and acetaminophen are retained by the cyclodextrin inclusion complex within the pharmaceutically acceptable carrier;
• and,
• admixing an enzyme with the cyclodextrin inclusion complex and the pharmaceutically acceptable carrier, the enzyme having a cyclodextrin-degrading activity capable of digesting the cyclodextrin retaining the cyclodextrin inclusion complex, wherein the cyclodextrin-degrading activity is activated on oral delivery of the composition to a human subject so as to release the N-acetylcysteine and acetaminophen from the cyclodextrin cavity.
