Abstract
As a method for producing a 3-hydroxyisobutyric acid ester using a biocatalyst, a method for producing a 3-hydroxyisobutyric acid ester, including a step of allowing an alcohol or phenol to act on 3-hydroxyisobutyryl-CoA in the presence of alcohol acyltransferase to produce a 3-hydroxyisobutyric acid ester is provided.
Claims9
A method for producing a 3-hydroxyisobutyric acid ester, comprising reacting an alcohol or phenol with 3-hydroxyisobutyryl-CoA in the presence of an alcohol acyltransferase to produce a 3-hydroxyisobutyric acid ester, wherein
- the alcohol acyltransferase is any one of the following proteins (1) to (3):
- (1) a protein comprising the amino acid sequence of SEQ ID NO:1;
- (2) a protein comprising an amino acid sequence derived from the amino acid sequence of the SEQ ID NO:1 by deletion, insertion, substitution and/or addition of one or several amino acids, and having an alcohol acyltransferase activity catalyzing esterification of 3-hydroxyisobutyryl-CoA;
- and
- (3) a protein comprising an amino acid sequence having a sequence identity of 80% or more with the amino acid sequence of SEQ ID NO:1 and having an alcohol acyltransferase activity that catalyzes esterification of 3-hydroxyisobutyryl-CoA,
- wherein the presence of alcohol acyltransferase is provided by a genetically modified microorganism expressing the alcohol acyltransferase.
A method for producing a methacrylic ester, comprising:
- reacting an alcohol with 3-hydroxyisobutyryl-CoA in the presence of alcohol acyltransferase to produce a 3-hydroxyisobutyric acid ester, and
- subjecting the 3-hydroxyisobutyric acid ester to a dehydration reaction to produce a methacrylic ester, wherein
- the alcohol acyltransferase is any one of the following proteins (1) to (3):
- (1) a protein comprising the amino acid sequence of SEQ ID NO:1;
- (2) a protein comprising an amino acid sequence derived from the amino acid sequence of SEQ ID NO:1 by deletion, insertion, substitution and/or addition of one or several amino acids, and having an alcohol acyltransferase activity catalyzing esterification of 3-hydroxyisobutyryl-CoA;
- and
- (3) a protein comprising an amino acid sequence having a sequence identity of 80% or more with the amino acid sequence of SEQ ID NO:1 and having an alcohol acyltransferase activity that catalyzes esterification of 3-hydroxyisobutyryl-CoA,
- wherein the presence of alcohol acyltransferase is provided by a genetically modified microorganism expressing the alcohol acyltransferase.
