Abstract
Methods, devices, and systems for improving the quality of electrospray ionization mass spectrometer (ESI-MS) data are described, as are methods, devices, and systems for achieving improved correlation between chemical separation data and mass spectrometry data.
Claims20
A method comprising:
- providing a sample comprising at least one analyte of interest;
- introducing the sample into a fluid channel;
- separating the sample within the fluid channel using isoelectric focusing, wherein said separation generates at least one resolved analyte peak;
- upon mobilization of the fluid channel's contents towards a fluid channel outlet calculating a velocity of at least one resolved analyte peak that was mobilized;
- using the calculated velocity to determine a time at which the resolved analyte peak that was mobilized reaches the fluid channel outlet;
- and
- wherein the time at which the resolved analyte peak that was mobilized reaches the fluid channel outlet is used to correlate mass spectrometer data with the resolved analyte peak after isoelectric focusing was completed and prior to mobilization.

A computer-implemented method comprising:
- receiving, using a processor, image data comprising two or more images acquired using a detector configured to image all or a portion of a fluid channel in a capillary or a microfluidic device;
- processing the image data using the same or a different processor to determine a position of a resolved analyte peak within the separation channel in the two or more images;
- calculating, using the same or a different processor, a velocity of the resolved analyte peak upon mobilization based on the position of the resolved analyte peak in the two or more images and a known time interval between acquisition of the two or more images;
- and
- determining, using the same or a different processor, using the calculated velocity, a time at which the resolved analyte peak will reach a fluid channel outlet;
- and
- correlating, using the same or a different processor, mass spectrometer data with the resolved analyte peak after isoelectric focusing was completed and prior to mobilization by using the time at which the resolved analyte peak reaches the fluid channel outlet.
