Abstract
Disclosed herein are antibodies that specifically bind to a methotrexate analyte. Such antibodies can be used to detect a methotrexate analyte in a sample, such as in a homogeneous enzyme immunoassay method. Hapten structures to elicit such antibodies and conjugates useful in immunoassays are also described.

A compound of Formula 1:
- wherein R1 is attached at the para or meta position;
- wherein:
- R1 is —C(O)NH(CH2CH2O)2(CH2)2NHC(O)CH2—Z or —C(O)NH(CH2)2NHC(O)CH2—Z,
- when R1 is attached at the para position;
- or
- R1 is —CH2NHC(O)(CH2)7NHC(O)CH2—Z;
- —CH2NHC(O)(CH2)2NHC(O)CH2—Z;
- —CH2NHC(O)(CH2)2O(CH2)4NHC(O)CH2—Z;
- or —CH2NHC(O)CH2—Z, when R1 is attached at the meta position;
- and
- Z is selected from the group consisting of hydrogen, SH, S-acyl, halogen, NH2, epoxy, maleimidyl, haloacetamide, activated carboxyl, azide, alkene, an immunogenic carrier, a protein, a label enzyme, a solid support and salts thereof.
