Abstract
Engineered promiscuous biotin ligases and methods of using them in proximity labeling are described. In particular, the invention provides novel biotin ligase variants having increased promiscuous biotinylation activity capable of proximity labeling of proteins in live cells in as little as 10 minutes.
Claims21
A modified biotin ligase comprising at least one mutation comprising an amino acid substitution selected from the group consisting of Q65P, M209V, V160A, S150G, L151P, I305V, I87V, T192A, K194I, E140K, Q141R, M241T, and S263P, wherein positions of the amino acids are numbered relative to the reference wild-type biotin ligase sequence of SEQ ID NO:7, and wherein the biotin ligase amino acid sequence is at least 90% identical to the sequence of SEQ ID NO:
- 7.
