Abstract
The present disclosure provides methods and systems for generation of compositions comprising two or more sets of molecules. Compositions herein may comprise sets of molecules of different types. Sets of molecules may be generated by attachment to supports. Different types of molecules may be used to generate a large number of unique molecules. One or more types of molecules may be used to identify one or more additional types of molecules. Compositions of the present disclosure may be used in, for example, nucleic acid sequencing.
Claims20
A method for generating a barcoded nucleic acid molecule, comprising:
- a) subjecting a B-cell and a support to conditions sufficient to permit said B-cell to bind to said support and generate a B-cell-support-complex comprising a nucleic acid barcode sequence, wherein:
- (i) said B-cell comprises a receptor;
- (ii) said support is coupled to a polypeptide and said nucleic acid barcode sequence;
- and (iii) said receptor binds to said polypeptide, thereby binding said B-cell to said support;
- b) partitioning said B cell-support-complex comprising said nucleic acid barcode sequence into a partition;
- and
- c) in said partition, using said nucleic acid barcode sequence to generate a barcoded nucleic acid molecule comprising (i) a nucleic acid sequence that encodes at least a portion of said receptor or reverse complement of said nucleic acid sequence and (ii) said nucleic acid barcode sequence or reverse complement of said nucleic acid barcode sequence.
