Abstract
The invention generally relates to methods for quantifying an amount of enzyme molecules. Systems and methods of the invention are provided for measuring an amount of target by forming a plurality of fluid partitions, a subset of which include the target, performing an enzyme-catalyzed reaction in the subset, and detecting the number of partitions in the subset. The amount of target can be determined based on the detected number.
Claims20
A method for detecting target molecules in a fluid sample, the method comprising:
- providing, to a microfluidic device, a fluid sample comprising a target molecule, a plurality of bead-linked reactive components for binding the target molecule, and a detection component for binding to the target molecule;
- binding the target molecule to one of the bead-linked reactive components and to the detection component;
- partitioning, in the microfluidic device, the fluid sample into plurality of separate and isolated partitions of fluid such that at least one of the partitions includes the target molecule bound to the detection component and the one bead-linked reactive component;
- and
- monitoring each of the plurality of partitions for detection of a detectable label released by the detection component bound to the target molecule.
