Abstract
Use of cyclosporin H (CsH) or a derivative thereof for increasing the efficiency of transduction of an isolated population of cells by a viral vector and/or increasing the efficiency of gene editing of an isolated population of cells when transduced by a viral vector.
Claims17
A method of transducing a population of cells comprising the steps of:
- (a) contacting the population of cells with cyclosporin H (CsH);
- and
- (b) transducing the population of cells with a viral vector, wherein the viral vector is a lentiviral or gamma-retroviral vector and wherein the viral vector is pseudotyped to enter cells via an endocytosis-dependent mechanism,
- wherein the population of cells comprises:
- (i) CD34+ HSPCs;
- (ii) CD14+ monocytes;
- (iii) CD4+ or CD3+ T cells;
- (iv) CD34+ bone marrow cells;
- or (v) CD34+ peripheral blood cells.
