Abstract
The present disclosure provides for recombinant oncolytic viruses with gene deletions or insertions which result in downregulation of Major Histocompatibility Complex class I and alternatively or additively upregulation of Major Histocompatibility Complex class II. Immunologic and pharmaceutical compositions comprising these recombinant viruses and methods of using these compositions are also presented.
Claims19
A composition, wherein the composition comprises:
- an oncolytic virus, wherein the oncolytic virus comprises a genome modification, wherein the genome modification comprises an exogenous nucleic acid encoding for a membrane associated extracellular matrix (ECM) degrading enzyme comprising an amino acid sequence having at least 98% sequence identity to SEQ ID NO:
- 30 and encoding for the following functional components:
- a bacterial hyaluronidase enzyme or a functional variant thereof, wherein the bacterial hyaluronidase enzyme is soluble in its naturally occurring form;
- an N-terminal secretory signal sequence;
- and
- a signal sequence for membrane association.
