Abstract
Disclosed are methods and systems for the analysis activity of enzyme disintegrin and metalloproteinase with a thrombospondin type 1 motif, member 13 (ADAMTS13) in a sample. The methods and systems disclosed herein can be useful for diagnosis of thrombotic thrombocytopenic purpura in a patient.
Claims29
A method for determining a disintegrin and metalloproteinase with a thrombospondin type 1 motif, member 13 (ADAMTS13) enzyme activity in a sample, comprising:
- (a) incubating the sample with an exogenous peptide substrate for ADAMTS13 under conditions allowing for enzymatic cleavage of the exogenous peptide substrate by ADAMTS13 to produce an enzymatic cleavage product, wherein the exogenous peptide substrate comprises an ADAMTS13 cleavage site, and wherein the enzymatic cleavage product comprises a sequence with at least 70% sequence identity to SEQ ID NO:4;
- (b) ionizing the enzymatic cleavage product to generate a multiply charged gas-phase ion of the enzymatic cleavage product;
- and,
- (c) analyzing the multiply charged gas-phase ion by tandem mass spectrometry to determine presence or amount of enzymatic cleavage product in the sample, wherein the presence or the amount of the enzymatic cleavage product in the sample is indicative of presence or amount of enzyme activity of ADAMTS13 in the sample.
