Acetylsalicylic acid derivative and application thereof

A pharmaceutical composition as a transdermal dosage form or a transmucosal dosage form, comprising a therapeutically effective amount of the compound represented by Formula (1):

• wherein R1 and R2 each independently represent H, unsubstituted C1-C30 alkyl, unsubstituted alkoxy, unsubstituted C2-C30 alkenyl, unsubstituted C6-C30 aryl, unsubstituted C7-C30 aralkyl, or unsubstituted C3-C30 cycloalkyl, or R1 and R2 form 4-, 5-, 6- or 7-membered heterocycle, together with the N to which they are linked;
• R3 represents unsubstituted C1-C12 alkyl;
• n is 1;
• and
• a crystal of the compound represented by Formula (1) comprises characteristic peaks in an X-ray powder diffraction (XRPD) pattern at the following 20 angles:
• 9.4°±0.2°, 10.3°±0.2°, 12.2°±0.2°, 18.1°±0.2°, 21.8°±0.2°, 24.4°±0.2°, 29.4°±0.2°, 30.6°±0.2°, 31.9°±0.2°, 36.6°±0.2°,
• or the pharmaceutically acceptable salt thereof, and a pharmaceutically acceptable carrier.

A method of treating acetylsalicylic acid indications or anti-platelet aggregation, comprising the step of applying the compound represented by Formula (1) or the pharmaceutically acceptable salt in a transdermal dosage form or a transmucosal dosage form to a subject to be treated:

• wherein R1 and R2 each independently represent H, unsubstituted C1-C30 alkyl, unsubstituted alkoxy, unsubstituted C2-C30 alkenyl, unsubstituted C6-C30 aryl, unsubstituted C7-C30 aralkyl, or unsubstituted C3-C30 cycloalkyl, or R1 and R2 form 4-, 5-, 6- or 7-membered heterocycle, together with the N to which they are linked;
• R3 represents unsubstituted C1-C12 alkyl;
• n is 1;
• and
• a crystal of the compound represented by Formula (1) comprises characteristic peaks in an X-ray powder diffraction (XRPD) pattern at the following 20 angles:
• 9.4°±0.2°, 10.3°±0.2°, 12.2°±0.2°, 18.1°±0.2°, 21.8°±0.2°, 24.4°±0.2°, 29.4°±0.2°, 30.6°±0.2°, 31.9°±0.2°, 36.6°±0.2°.
