Abstract
Provided herein are methods, compositions and kits to extract and relatively enrich by physical separation or amplification short base pair nucleic acid in the presence of a high background of genomic material (e.g., host or maternal nucleic acids).
Claims17
A kit for extracting target nucleic acid comprising:
- a) a set of solid supports consisting of a first solid support capable of reversibly binding nucleic acid and a second solid support capable of reversibly binding nucleic acid;
- b) a set of binding buffers consisting of a first binding buffer and a second binding buffer, wherein:
- the first binding buffer is formulated to comprise guanidine (iso)thiocyanate, wherein the guanidine (iso)thiocyanate is present at a concentration of 2 M to 4 M, and
- the second binding buffer is formulated to comprise guanidine (iso)thiocyanate, wherein the guanidine (iso)thiocyanate is present at a concentration above 4 M;
- c) a set of wash buffers consisting of a first wash buffer, a second wash buffer, and a third wash buffer;
- and
- d) instructions for performing the target nucleic acid extraction.
