Abstract
The present disclosure relates to the production of cannabinoids in yeast. In as aspect there is provided a genetically modified yeast comprising: one or more GPP producing genes and optionally, one or more GPP pathway genes; two or more olivetolic acid producing genes; one or more cannabinoid precursor or cannabinoid producing genes; one or more Hexanoyl-CoA producing genes, and at least 5% dry weight of fatty acids or fats.
Claims10
A genetically modified yeast comprising:
- (a) a first polynucleotide encoding a first polypeptide having at least 70% amino acid sequence identity to SEQ ID NO:4, wherein said first polynucleotide has farnesyl diphosphate synthase (ERG20) activity;
- (b) a second polynucleotide encoding a second polypeptide having at least 70% amino acid sequence identity to SEQ ID NO:7, wherein said second polypeptide has olivetol synthase (OLS) activity;
- (c) a third polynucleotide encoding a third polypeptide having at least 70% amino acid sequence identity to SEQ ID NO:8, wherein said third polypeptide has olivetolic acid cyclase (OAC) activity;
- (d) a fourth polynucleotide encoding a fourth polypeptide having at least 70% amino acid sequence identity to SEQ ID NO:12, wherein said fourth polypeptide has soluble aromatic prenyltransferase (NphB) activity;
- and
- (e) at least 5% dry weight of fatty acids or fats.
