Abstract
The present disclosure provides methods of generating supports (e.g., beads) comprising barcode molecules coupled thereto. A barcode molecule coupled to a support may comprise a barcode sequence and a functional sequence. A barcode molecule may be generated using two or more ligation reactions in a combinatorial fashion. A support comprising two or more different barcode molecules may be useful for analyzing or processing one or more analytes such as nucleic acid molecules, proteins, and/or perturbation agents.
Claims18
A method for generating nucleic acid barcode molecules, comprising:
- (a) providing a plurality of nucleic acid molecules coupled to a support;
- (b) coupling one or more additional nucleic acid molecules to nucleic acid molecules of the plurality of nucleic acid molecules coupled to the support, thereby assembling a plurality of nucleic acid barcode molecules coupled to the support, wherein the plurality of nucleic acid barcode molecules comprises a plurality of target products and a plurality of off-target products;
- and
- (c) digesting off-target products of the plurality of off-target products with an exonuclease, wherein target products of the plurality of target products comprise an exonuclease-resistant bond, and wherein fewer than 50% of the target products comprising the exonuclease-resistant bond are digested with the exonuclease.
