Abstract
An automatic focus system for an optical microscope that facilitates faster focusing by using at least two offset focusing cameras. Each offset focusing camera can be positioned on a different side of an image forming conjugate plane so that their sharpness curves intersect at the image forming conjugate plane. Focus of a specimen can be adjusted by using sharpness values determined from images taken by the offset focusing cameras.
Claims11
A system for automatically focusing a microscope, comprising:
- a stage for positioning a specimen on a first image forming conjugate plane;
- an illumination unit comprising:
- a primary illumination source configured to emit light in a first wavelength range,
- a secondary illumination source configured to emit light in a second wavelength range,
- a focusing mechanism positioned in a light path emanating from the secondary illumination source, the focusing mechanism configured to project a focusing pattern on the specimen, and
- one of:
- a dichroic configured to reflect light in the second wavelength range and allow light in the first wavelength range, or
- a band filter configured to transmit light in the first wavelength range and block light in the second wavelength range;
- a focusing unit configured to focus the microscope, the focusing unit comprising a first focusing camera positioned at a first offset distance from a second image forming conjugate plane;
- and
- a control unit in communication with the focusing unit, the control unit configured to determine when the specimen is in focus.
A method for automatically focusing a microscope, comprising:
- setting a first focusing camera at a first offset distance from an image forming conjugate plane;
- setting a second focusing camera at a second offset distance from the image forming conjugate plane;
- illuminating a specimen positioned on a stage at a second image forming conjugate plane with at least one of a primary illumination source or a secondary illumination source, the primary illumination source configured to emit light in a first wavelength range, the secondary illumination source configured to emit light a second wavelength range, wherein illuminating the specimen comprises:
- transmitting a first emitted beam from the primary illumination source to the specimen using a dichroic, the dichroic configured to allow light in the first wavelength range, and
- reflecting a second emitted beam from the secondary illumination source using the dichroic, the dichroic further configured to reflect light in the second wavelength range;
- and
- determining that the specimen is in focus when a sharpness value of the specimen using the first focusing camera is within a threshold range of a second sharpness value of the specimen using the second focusing camera.
A system for automatically focusing a microscope, comprising:
- an illumination unit comprising:
- a primary illumination source,
- a secondary illumination source, and
- a focusing mechanism positioned in a light path emanating from the secondary illumination source, the focusing mechanism configured to project a focusing pattern on a specimen;
- a focusing unit for focusing the microscope, the focusing unit comprising a first focusing camera positioned at a first offset distance from an image forming conjugate plane and a second focusing camera positioned at a second offset distance from the image forming conjugate plane;
- and
- a control unit in communication with the focusing unit, the control unit configured to determine when the specimen is in focus, wherein the control unit determines that the specimen is in focus when a sharpness value of the specimen using the first focusing camera is within a threshold range of a second sharpness value of the specimen using the second focusing camera.
A method for automatically focusing a microscope, comprising:
- setting a first focusing camera at a first offset distance from an image forming conjugate plane;
- setting a second focusing camera at a second offset distance from the image forming conjugate plane;
- transmitting a first emitted beam from the primary illumination source to the specimen using a band filter, the band filter configured to transmit light in the first wavelength range,
- blocking a second emitted beam from the secondary illumination source using the band filter, the band filter further configured to reflect light in the second wavelength range, and
- allowing a portion of the second emitted beam from the secondary illumination source using the band filter, the portion of the second emitted beam being location within a region of a focusing mechanism;
- and
- determining that the specimen is in focus when a sharpness value of the specimen using the first focusing camera is within a threshold range of a second sharpness value of the specimen using the second focusing camera.
