Abstract
In some aspects, the disclosure relates to methods for improving titer and yield of viral vector production. In some embodiments, the methods comprise transient silencing of transgene expression during packaging of a viral vector.
Claims11
A method for controlling recombinant lentivirus production yield, the method comprising:
- (i) introducing into a mammalian cell a lentiviral transfer plasmid comprising a transgene encoding an RNA transcript comprising one or more non-mammalian artificial miRNA (amiRNA) binding sites;
- (ii) introducing into the mammalian cell a nucleic acid capable of expressing a non-mammalian amiRNA that specifically binds to the one or more non-mammalian amiRNA binding sites and inhibits translation of the RNA transcript of the transgene;
- and
- (iii) replicating the lentiviral transfer plasmid within the host cell.
