Abstract
The present disclosure relates to genetically modified non-obese diabetic (NOD) mice deficient in murine class I MHC molecules, class II molecules, or both class I and class II MHC molecules. The MHC knockout transgenic mice provided herein are useful, for example, for developing therapies for diabetes.
Claims9
A method comprising:
- (a) providing a non-obese diabetic (NOD) mouse having a NOD/ShiLtJ or NOD/ShiLtDvs genetic background,
- wherein (i) the mouse is homozygous for a knockout mutation in a H2-K1d allele, the mouse is homozygous for a knockout mutation in a H2-D1b allele, (iii) murine H2-K and H2-D MHC class I molecules are not expressed in the mouse, and (iv) the mouse is resistant to type 1 diabetes and insulitis out to thirty weeks of age;
- (b) administering a test agent to the mouse;
- and
- (c) assaying the mouse for a symptom of diabetes.
